In my opinion this request should not be granted due to the fact you mention
the part you want to add is a spare part that Lego provides as a replacement.
This means, correct me if I am wrong, this part does not belong to the set but
Lego only offers it to you because they do not make the other part and the new
part has the same shape and use.

So, if I understand your request well, you did not have the tile for this set,
you went to Lego and they provided you with this part. Therefore it never belonged
to the set.

If I am not correct in understanding your request and you mean something different
like you found this round tile in a new and sealed set than it is a different
matter.

The LEGO replacement parts website only lists the current version of parts in
production that can be used to replace older versions of parts that are no longer
in production.

In this case, the set was produced with the older version of the part:
 
Part No: 4150  Name: Tile, Round 2 x 2

Parts: Tile, Round {White}

The newer version of the part
 
Part No: 14769  Name: Tile, Round 2 x 2 with Bottom Stud Holder
* 
14769 Tile, Round 2 x 2 with Bottom Stud Holder
Parts: Tile, Round {White}
was released in 2013 and would never have been included in this set.

Only different versions of parts that were *actually* included in a set during
the production run of a set are able to be added as Alternate Items to a set's
inventory.
