A brick and mortar store is an actual physical store (like in a mall) compared
to the BrickLink stores which are virtual.

They asked you in essence if you had an actual store they could visit in person.
They may have been hoping for a free Hold for Pick-Up shipping option.

Actually, a good question...

And nothing to do with “virtual” as commonly characterized hereabouts. All things
are virtual in the human experience. Online or brick n’mortar, both versions
sell and convey the same physical item or product. Share an end goal.

That said, in contemporary parlance “brick and mortar” denotes a physical location
that one can present oneself anonymously. Visit, in other words. With online
stores, that’s not possible unless you’re willing to ponder whether or not an
on-line store that accepts visits or local pick-ups, is then made “Brick and
Mortar” by definition? I think not.
