I have a Wanted List and all items have the condition new.
When I'm now trying to buy all the items it seems like not all are available
in 'new' condition. So I I'd like to switch the condition for some
items to 'used' during the buying. Is this possible?
One possibility would be to open the Wanted List in the Bricklink App and change
all the conditions of the items that are left to used, save it and upload it
on the website? Is there an easier way?
Or another Wanted List question. When all items have the condition 'any',
are new parts preferred or how does this work?

Not “on the fly” / during the buying.

You can open your WL,
(set the number of items to 100 or more (you can edit the URL) to have all of
them on one page),
then select them all (checkbox in the header, ticking it ticks all the items
on the page),
then you click the Condition button and follow fill the form.

If you use Auto-select / EasyBuy, it'd generally be the cheapest one that's
chosen.
If you use the WL in a store, all the lots are shown.
What I'd do in a case like this would be to have two lists: one with New
and one with Used.
Try to buy the New one, and then, with the Wanted Items tab, see which ones weren't
found and unselect them and select their counterparts in the Used WL.

Because of about 100 unique lots missing it would takes a lot of time. But I
also don't see a better approach...

Note that if a part only exists in Used, it may be a good idea to think about
it more deeply.
Well, unless it's a stickered part: stickered parts are necessarily and obviously
Used (otherwise, you want the plain parts and the sticker sheets).
If it's for completing old sets with old parts, then it's logical if
some parts only exist Used. Otherwise, if it's for a MOC, then it may be
a better idea to modify the build to use more modern and common parts or colours.
Also, if the parts only exist in Used, they may be expensive and/or rare and
it may be a better idea to find stores for them first.
(So, first identify these parts and then search for the stores. If you do it
in one go, you'll need more stores and more shipping…. Always a good idea
to search for stores for the rarer parts first.)
It also depends on the time and money you want to spend.
